Applications:

Measurement of masses of intact proteins for quality control. Gel-purified protein identification by peptide mass fingerprinting and peptide fragmentation. De novo sequencing of peptides for protein identification. Quantification of differential protein expression by gel-free proteomics and 2D-LC-MS. Identification of sites of covalent modification by collision-induced dissociation (CID) fragmentation of peptides or by electron transfer dissociation of peptides (ETD).

Equipment:

ThermoScientific LTQ XL electrospray ionization linear ion trap mass spectrometer fitted with CID and ETD fragmentation with possibilities of performing nano-electrospray ionization analysis or peptide analysis by coupling to a Dionex capillary HPLC system for on-line 1D- or 2D-LC-MS. Bioinformatics workstation for protein identification and label-free quantification: Proteome Discoverer, BioWorks, Sieve, Biomass, Sequest.

Use and conditions:

- Open to all members of the de Duve Institute (contact Didier Vertommen for discussing experimental strategy and calendar).
- External scientists are welcome to contact Didier Vertommen for conditions.
- Analyses can be performed within the framework of a scientific collaboration or as a service.
- Participation towards costs of sample preparation and running costs. Contact Didier Vertommen for price list.
